The 20-year-old produced a flawless round that included 7 birdies and no bogeys to hold a one shot advantage over Australian Su Oh at the Sime Darby LPGA in Kuala Lumpur.
A trio of South Korea's Eun-Hee Ji, China's Shanshan Feng and Sweden's Madelene Sagstrom are one shot further back on 66.
World No.8 Ko is chasing her first tournament win in over a year and says she feels that her form is coming back.
"I was struggling a little bit during the middle of my season this year," said Ko.
"I played great in Indiannapolis - I didn't end up winning but my second place finish gave me confidence."
"Week in week out the talent level on the tour doesn't change much, but the person who is confident and comfortable and positive, they are the players who are going to keep consistently playing well."
Ko said she is looking to maintain her new found form over the next couple of days in Kuala Lumpur.
"The more I see the puts go in or the more I see the under par rounds, the more confident I feel going into the next round."
So hopefully I will play with confidence in the next few days."
